What do you think are the main differences between having a do follow link and a no follow link?

And if you have a website or a blog, will you enable dofollow for the comments, if not why?

Imo, I guess if you are a new blogger, better enable dofollow for comments. Sure you will get lots of spam. But along with that you will get valid/engaging comments from other fellow newbie bloggers, who are interested in getting a dofollow backlink. That way the chances are like you get a comment, a valid trusted link + a frequent visitor. And google do love websites with a lot of comments.

For a new blog, I think enabling do-follow comments is a must! Even though you'll find a lot of comments are just there for the link, it still encourages others to comment on your blog which increases your community participation :) There are some who say that having too many outbound links is bad for SEO though... :(

If the website have a good number of inbound links/inner links(the links within the webpages -What do we call them? :p) the number of outbound links doesn't matter. I think I read this in some article. Not sure though! :)

Well I don't know, if you get too many outbound links not related to your site, Google can see your site as Spammy and that will affect SEO... But generally you shouldn't have to worry about it :)

Yeah thats for sure. You have to be careful about that. The links from your website should always be related to your website or at least related to the topic on which you get the comment on!
